1月24日,上海科技大學iHuman研究所饒子和院士與楊海濤教授的合作團隊在Cell上發表了題為Crystal Structures of Membrane Transporter MmpL3, an Anti-TB Drug Target的研究論文,文中解析了結核分枝桿菌(M . smegmatis)MmpL3完整的晶體結構以及和四個潛在藥物形成的復合物的晶體結構。MmpL3結構的解析,對促進MmpL3的抑制劑的篩選以及抗結核病藥物的研究具有極其重要的意義。
提到恥垢分枝桿菌(Mycobacterium Smegmatis),此前BioArt也報道過饒子和院士團隊2018年10月份發表在Science上的題為An electron transfer path connects subunits of a mycobacterial respiratory supercomplex的文章【12】(Science丨立足抗結核病藥物靶點開發,饒子和院士團隊歷時多年解析分歧桿菌呼吸體超復合物III2IV2SOD2原子分辨率結構)。文中解析了恥垢分枝桿菌呼吸體超復合物III2IV2SOD2原子分辨率冷凍電鏡結構。
M . smegmatis是一種過氧化氫耐受株。與結核分枝桿菌(M.tuberculosi)比較接近,同屬分枝桿菌屬。恥垢分枝桿菌相對結核桿菌操作安全性要好的多,因此可作為研究結核桿菌的理想替代研究對象。饒子和院士團隊多年來專注于分枝桿菌編碼的關鍵蛋白分子的結構生物學研究,此次解析的重要膜蛋白分子MmpL3的晶體結構,也是多年來厚積薄發的結果。
上海科技大學iHuman研究所劉志杰教授課題組長期聚焦大麻素受體的系統性研究,試圖理解其調控的重要生理功能和其結構基礎。目前他們已分別發表了CB1與拮抗劑AM6538復合物的晶體結構(Cell, 2016)、CB1分別與激動劑AM841及AM11542復合物的晶體結構(Nature, 2017)。在前期工作的基礎上,為攻克CB2受體結構及調控機制,劉志杰團隊與美國東北大學的Alexandros Makriyannis教授團隊合作并設計了CB2的新型拮抗劑AM10257。研究團隊通過理性突變位點預測和大量實驗篩選,最終獲得了高質量的CB2蛋白樣品并成功解析了CB2與AM10257復合物2.8埃分辨率的晶體結構。相關工作以Crystal Structure of the Human Cannabinoid Receptor CB2為題于2019年1月11日在線發表于Cell雜志上,該論文1月24與饒子和院士團隊的Cell論文同時正式出版。
