iTRAQ(Isobaric Tag for Relative Absolute Quantitation)和TMT(Tandem Mass Tags)技術是分別由美國AB Sciex公司和Thermo公司研發的多肽體外標記定量技術。這兩種技術采用2-10種穩定同位素標簽，通過特異性 標記多肽的氨基基團，然后進行串聯質譜分析，可同時比較多達10種不同樣本中蛋白質的相對含量。
iTRAQ/TMT試劑由三部分組成：報告基團、平衡基團和反應基團。反應基團形成2-10種等質量同位素標簽。iTRAQ/TMT試劑標記酶解后的肽段，可與氨基酸N端及側鏈的伯胺基團發生反應。在一級譜圖中，來自于不同樣本的同一肽段，標記后表現為相同的質荷比。在二級譜圖中，報告基團、平衡基團和反應基團之間的鍵斷裂，帶不同同位素標簽的同一肽段產生質量不同的報告離子，根據報告離子的豐度可獲得樣本間相同肽段的定量信息，再經過軟件處理得到蛋白質的定量信息。其基本原理與技術流程如下圖所示：
